Numerous methods for exosome purification, such as ultracentrifugation and microfiltration, size exclusion chromatography (SEC), and immunoaffinity capture, have been developed to improve the concentration and quality of exosomes.
Microfiltration and Ultracentrifugation
The most widely used technique for isolating extracellular vesicles is ultracentrifugation. Using this method, vesicles are separated from other sample components based on the rates at which they sediment, which are primarily affected by size and density. Exosomes can be easily isolated from samples using the isolation/enrichment technique known as microfiltration, which uses a hydrophilized polyvinylidene difluoride membrane to do so with less co-purification of plentiful non-exosomal proteins.
